RESEARCH PROJECTS

  • Postdoctoral project: Transforming growth factor-β (TGF-β) exerts a context-dependent influence on cancer, acting as a tumor suppressor in early disease but promoting invasion, therapy resistance, and immune evasion in advanced glioblastoma (GBM). To better understand its pro-tumorigenic role, this study examined how TGF-β regulates cytoskeletal remodeling and epithelial-to-mesenchymal transition (EMT) in GBM cells through Smad/β-catenin signaling and suppression of the tumor-modulating protein Testin (TES). Using LN229 and U87 cell lines, we observed that TGF-β treatment drives a dose-dependent increase in cell migration with concomitant loss of adhesion. These functional changes aligned with pronounced morphological alterations, including cell elongation and F-actin reorganization, characteristic of EMT. Molecular analyses further showed activation of canonical Smad and non-canonical β-catenin pathways, along with significant downregulation of Testin. Preliminary evidence suggests that this reduction may arise from TGF-β-induced promoter hypermethylation. Collectively, the data support a model in which TGF-β enhances GBM aggressiveness by simultaneously activating pro-invasive signaling pathways and epigenetically suppressing Testin. Targeting this integrated TGF-β/Smad/β-catenin/Testin axis—either by restoring Testin expression or inhibiting upstream effectors—may offer a promising strategy to limit GBM invasion and improve responsiveness to chemotherapy.
  • Collaborative project: The development of nanoparticle-based bio-imaging systems offers a powerful strategy for improving glioblastoma (GBM) detection and monitoring. Engineered nanoparticles provide enhanced contrast, targeted delivery, and stable signal retention within the tumor microenvironment. Surface functionalization with ligands enables selective binding to GBM-specific markers, improving imaging accuracy while reducing off-target effects. These platforms support fluorescence imaging—allowing comprehensive visualization of tumor boundaries, invasion fronts, and treatment response in vitro. By integrating high sensitivity with molecular specificity, nanoparticle-based imaging systems hold strong potential to overcome current limitations in GBM diagnosis and real-time therapeutic assessment.
  • Ph.D. Project: Project involved investigating the paracrine interaction between the breast cancer cell and stroma in the Tumor microenvironment (TME). In this study, using the 3D co-culture model it was demonstrated, bone marrow-derived mesenchymal stem cells (bmMSCs) induce proliferation, alters cytoskeletal organisation, promote epithelial-mesenchymal transition (EMT) and invasion in ductal carcinomain situ (DCIS)-derived breast cancer cell lines and patient-derived xenograft (PDX) samples of breast cancer. Since bm-MSCs participate in the transformation of DCIS into invasive breast cancer (IBC), further investigation is require to identify a potential targeted-therapeutic to stop breast cancer progression, studying through the MSC-exerted paracrine signalling along with the juxtacrine and exosomal signalling axes in breast cancer in the 3D co-culture paradigm.
