Project title: "Conception, development, and characterization of a novel topical formulation for the treatment of plaque psoriasis" The objective of the study project was to develop a new topical delivery system in order to provide the loco-regional release of fixed-dose combination drugs: Vit-D analogue and corticosteroids into the skin, and to reduce the systemic absorption and side effects of the drugs The drugs used in the combination in order to have a synergistic effect and acting from two different mode of actions. The ingredients were screened and selected after optimal results. FDC drugs loaded poly (D,Lactic coglycolic acid) PLGA micro particles were prepared by solvent evaporation technique. The analytical methods- UV and HPLC was developed for analysis of drugs. Particle size analysis and morphological characterization was done by Optical microscopy, SEM, FTIR, DSC. In addition project also involved the development of novel modern base "Bigel" using some specific natural ingredients and edible oils indicated in the Unani and Ayurvedic system of medicine for the treatment of plaque psoriasis. The developed topical formulation base was tested for Physical evaluation (color, odor, texture, phase separation, oil leakage), rheological parameters and texture analysis (hardness, stickiness and cohesiveness) through Optical Microscopy, Homogenizer, Freeze thaw procedure, Texture analyzer, Rheometer etc in order to obtain the best optimized Bigel base. The in-vitro drug release and permeability of the final topical formulation was accessed through confocal microscopy using guinea pig skin. The final formulation comprising FDC drugs loaded PLGA micro particles in Bigel base was subjected to animal studies for amelioration of Psoriasis symptoms and results were compared with the marketed formulation.
