Design and Expression of humanized antibodies against soluble Interleukin-6 receptor (sIL-6R) and soluble glycoprotein 130 (sGP130) in bacteria and animal cell lines for the treatment of Rheumatoid Arthritis Contents: Cloning, expression and purification of soluble IL-6 receptor, IL-6, soluble gp130 in animal cell lines and bacteria, development of collagen-induced arthritic models, gene expression studies using RT-PCR, affinity purification of recombinant proteins, production and purification of monoclonal and polyclonal antibodies, in-vivo and in-vitro functional studies and humanization of the selected antibodies against sIL-6R and sGP130.
2. Design and recombinant expression of EBOV-GP (Ebola glycoprotein), Protein A, G and L and preparation of affinity columns for purification of monoclonal antibody for diagnostic use
Contents: Cloning, expression and purification of EBOV-GP, Protein A, G and L in E.coli. System, generation of affinity columns using various coupling strategies for purification of monoclonal and polyclonal antibodies.
3. Development of hybrid vectors with intein-based dual tags
Contents: Preparation of chitin binding domains with intein based tags using Gibson Assembly methodology.
