Synthesis And Study Of Dihydropyrimidinone Derivatives Project is based on the synthesis of dihydropyrimidinone derivatives using Biginelli Reaction and evaluation of their antibacterial activity. The main goal of this project is to synthesize the new dihydropyrimidinone derivatives using different aldehydes, urea/thiourea and ethyl acetoacetate using suitable catalysts by biginelli reaction. The dihydropyrimidinones synthesized by biginelli reaction is made to react with sodium hypochlorite, sodium hydroxide and ammonium hydroxide. The derivatives obtained after treating with above components are heated with benzoylchloride, sodium acetate at 160c to afford the oxadiazolopyrimidine derivatives.cThe oxadiazolo derivatives that are prepared by using different aldehydes with urea and thiourea along with ethyl acetoacetate in presence of suitable catalyst are evaluated for their antimicrobial activity (antibacterial activity) using E.coli and B.subtilis. The oxadiazolopyrimidine derivatives prepared using the thiourea and aldehydes have more antimicrobial activity when compared to those prepared with urea. 2. Method Development and Validation of Fexofenadine Hydrochloride and Montelukast Sodium in Combine Tablet Dosage Form by RP-HPLC A simple, specific, accurate, and precise reverse phase High Performance Liquid Chromatographic (RP-HPLC) method was developed and validated for the estimation of Fexofenadine Hydrochlorid and Montelukast Sodium in pharmaceutical dosage form. A Waters Xterra 20µ column having 150 x 4.6 mm i.d. in isocratic mode, with mobile phase containing Mixed phosphate buffer of pH 6.8 and Acetonitrile(40: 60 v / v) was used. The flow rate was 0.8 ml / min and effluents were monitored at 226 nm. Chromatogram showed the main peak of Fexofenadine Hydrochloride at a retention time of 2.382 min and Montelukast Sodium at a retention time of 5.446. The method was validated for linearity, accuracy, precision, and limit of detection, limit of quantitation, robustnessand ruggedness. The limit of detection for estimation of Fexofenadine Hydrochloride and Montelukast Sodium was found to be 0.8175µg/ml and 0.1279µg/ml, respectively. The limit of quantitation for estimation of Fexofenadine Hydrochloride and Montelukast Sodium was found to be 2.4773µg/ml and 0.3876µg/ml, respectively. Recovery of Fexofenadine Hydrochloridewas found to be in the range of 99.20-100.22 % and Montelukast Sodium was found to be in the range of 99.41-9971 %.
