Ph.D. Thesis

  • Title - Identification and characterization of functional circular RNAs in skeletal muscle.
  • Supervisor- Dr. Amaresh C. Panda
  • Research Area- Circular RNAs (circRNAs) are produced by back-splicing, belong to the non-coding RNA class, and are known to regulate gene expression by interacting with miRNAs and RNA binding proteins (RBPs). We have identified hundreds of functional circRNAs expressed in aging mice skeletal muscle and C2C12 skeletal muscle cell line by high-throughput RNA sequencing. Identification of one abundant circRNA, circNfix, in mice skeletal muscle is found to regulate muscle differentiation by promoting Mef2c expression through sponging of miR-204-5p. Further, circRNA sequencing in C2C12 identified differentially expressed circCebref as a positive regulator of myotube formation by interacting with miR-181-5p and upregulating the expression of its downstream mRNA target, Pofut1.
